Thioacetic acid (ethanethioic acid) is a colorless or pale yellow liquid with a strong pungent odor, one of a key low-molecular-weight thiocarboxylic acids, widely used as a thioacylation reagent and sulfur source in organic synthesis. The product is soluble in water, ethanol, and ether.
Thioacetic acid can undergo radical addition with alkenes (to give thioesters), nucleophilic substitution with alkyl halides, reaction with acyl chlorides to give mixed thioanhydrides, and reaction with amines to give thioamides. It is also a commonly used thiol-protecting-group reagent (introduced via the acetylthio group) and can participate in electrophilic sulfuration reactions to introduce a sulfur atom into a target molecule.
Applications: Thioacylation reagent and sulfur source for thiol-bearing drugs, thioester prodrugs and thionucleosides; rubber vulcanization; thioxanthone photoinitiator synthesis.
