4-Fluorobenzoyl chloride is a fluorinated aromatic acyl chloride bearing a para fluorine and an acyl chloride group; its identity as a “fluorinated acylating reagent” is distinctive within the acyl-chloride family. It is a colorless to pale yellow liquid at room temperature with a boiling point of about 193℃, soluble in aprotic solvents, and reacts vigorously with water or alcohols.
The core advantage of 4-fluorobenzoyl chloride lies in the dual contribution of the fluorine atom: its strong electron-withdrawing effect significantly enhances the electrophilic reactivity of the acyl chloride, making acylation fast and efficient, while the small size of fluorine allows the acylated product to retain good biomembrane permeability.
